1. Prepare the plant tissue culture medium as stated in topic 3.

Plant Tissue Culturing (Initial Culture)

1. Cut the sucker to get the true shoot and sliced the true shoot into half.
2. Place the sliced shoot in the culture bottles with plant tissue culture medium.
3. Seal and label.

For subculturing

1. Remove the banana plantlets in the bottles.
2. Clean and cut the shoots.
3. Transfer the shoots into the prepared plant tissue culture medium.
4. Subculture up to 9 times. If intended for mass propagation, subculturing can be done up to 12 times.
