• Prepare the necessary chemicals needed for plant tissue culture. Make a stock solution.
• Weigh the solid chemicals like agar, sugar and growth hormone. Get a volume of the prepared stock solution for macronutrients, micronutrients and vitamins.
• Mix the solid and liquid components. Volume to the desired level with distilled water.
• Boil the solution for 15 minutes.
• Transfer the boiled solutions to the prepared bottles at the desired volume of the plant tissue culture medium.
• Sterilized the bottles with the plant tissue culture medium in the autoclave for 15 minutes at 15 psi.
• Transfer the bottles to the culture room.

The bottles with the plant tissue culture medium are now ready for inoculation
